PPQ Objectives
A Process Performance Qualification protocol must clearly articulate what the qualification is intended to demonstrate, namely that the commercial manufacturing process, executed at commercial scale using the finalized formulation and equipment, reliably produces product meeting all predetermined quality attributes across the range of normal process variability the facility will actually encounter in routine production. This objective statement should be specific to the actual product and process rather than a generic restatement of process validation's general purpose, since a clearly and specifically articulated objective directly shapes every subsequent protocol design decision, from what parameters and attributes to monitor through what acceptance criteria genuinely demonstrate that objective has been met.
Sampling and Monitoring Plan
The PPQ sampling plan should specify enhanced sampling and monitoring beyond routine commercial production levels, targeting both critical process parameters and critical quality attributes at a frequency and rigor sufficient to provide genuine statistical confidence in process performance, not merely confirm each individual batch happens to pass its own release testing. Sampling plan design should be informed by the process understanding developed during Stage 1, discussed in the related process validation lifecycle article, ensuring PPQ monitoring specifically targets the parameters and attributes already identified as genuinely critical rather than a generic, undifferentiated sampling approach applied uniformly across every conceivable parameter regardless of its actual demonstrated criticality.
Number of Batches
The number of PPQ batches should be statistically and risk-based justified, considering the process's inherent complexity, prior development-scale experience, and the level of confidence the specific number of batches will actually provide regarding genuine process consistency and capability. A commonly referenced convention of three PPQ batches should not be adopted reflexively without genuine justification specific to the process at hand, since a more complex or less well-characterized process may genuinely warrant additional batches to establish adequate confidence, while sufficient prior justified evidence might, in some circumstances, support a different number with appropriate scientific rationale.
Acceptance Criteria
PPQ acceptance criteria should address both individual batch release specification compliance and broader process capability and consistency across the complete set of PPQ batches, since the qualification's genuine purpose is demonstrating reliable, reproducible process performance, not merely confirming a small number of individual batches happened to pass their own release testing. Acceptance criteria should be predefined and documented in the approved protocol before PPQ batches are actually manufactured, preventing any temptation to adjust criteria after the fact based on whatever results the PPQ batches happen to produce.
Equipment and Facility Readiness Confirmation
Before PPQ batches begin, the protocol should confirm all supporting facility, utility, and equipment qualification has been successfully completed, since PPQ is intended to qualify the process itself operating within an already-qualified physical infrastructure, not to simultaneously qualify both the underlying infrastructure and the process running within it. Attempting to run PPQ batches before supporting equipment and facility qualification is genuinely complete risks confounding process-related findings with underlying infrastructure issues that should have been resolved during earlier qualification stages.
Personnel and Procedure Readiness
PPQ batches should be manufactured using finalized, approved batch records and by personnel representative of those who will actually perform routine commercial production, rather than an unusually experienced or specially selected team not representative of genuine ongoing commercial operations. Using non-representative personnel or draft, not-yet-finalized procedures during PPQ risks the qualification demonstrating a level of process performance not genuinely achievable once routine commercial production, with its more representative personnel and finalized procedures, actually begins.
Handling a PPQ Deviation or Failure
Any deviation occurring during PPQ batch manufacture should be thoroughly investigated following the same rigorous principles applied to any other deviation, with the investigation's conclusion, including whether the deviation reflects a genuine process robustness gap or an isolated, adequately explained occurrence, directly informing whether the affected PPQ batch can still support the overall qualification conclusion. A PPQ batch failure generally requires root cause investigation and corrective action before qualification can proceed, and depending on the nature and significance of the finding, may require repeating some or all PPQ batches following implementation of appropriate corrective action.
Transitioning From PPQ to Routine Commercial Production
Successful PPQ completion, documented in a formal report connecting the predefined objectives and acceptance criteria to the actual results obtained, supports transition to routine commercial production, at which point Stage 3 continued process verification, discussed in the related process validation lifecycle article, becomes the ongoing mechanism for confirming the process remains in this newly qualified, validated state throughout its full commercial life going forward.
